| Shrnutí: | Prostanoids are important signalling molecules which are produced locally both physiologically and in inflammatory diseases and have potent effects on a number of inflammatory processes. Prostanoids have often been the forgotten mediators of airway inflammation in the lung mainly because aspirin, an inhibitor of prostaglandin synthesis which has been used in medicine for almost a century, has little effect on airway function when given orally to the majority of asthmatics. Recent research in several areas has renewed interest in the role of prostanoids in airways inflammation. These have included studies showing that exogenously applied PGE sub(2) potently protects against several bronchoconstrictor challenges, studies showing that COX-2 the inducible form of cyclo-oxygenase is upregulated by the proinflammatory cytokines and mediators present in asthmatic airways and studies showing that inhaled cyclo-oxygenase inhibitors have protective effects on induced bronchoconstriction in asthma. The role of cyclo-oxygenase products in regulating airway function and the COX balance in asthma will be reviewed here. |
