| Description: | Non-coding RNAs (ncRNAs) are key genomic regulators, but their functions remain poorly understood. This dissertation used experimental and computational approaches to explore the non-coding genome. The study characterized the X-linked lncRNAs Crossfirre, Firre, and Dxz4 using loss-of-function in vivo models and revealed roles in autosomal gene regulation and sex-specific phenotypes. Additionally, the project developed a computational framework to predict the target genes of ncRNAs and identified 397 ncRNA-to-target links in mice and 2,291 in humans, along with their mode-of-action. ; Nicht-kodierende RNAs (ncRNAs) sind wichtige genomische Regulatoren, deren Funktionen wenig verstanden sind. In dieser Dissertation wurden experimentelle und computergestützte Ansätze verwendet, um das nicht-kodierende Genom zu untersuchen. Die Studie charakterisierte die X-chromosomalen lncRNAs Crossfirre, Firre und Dxz4 mit Hilfe von in vivo Modellen und zeigte Funktionen in der Regulation autosomaler Gene sowie geschlechtsspezifische Phänotypen. Zusätzlich wurde ein bioinformatisches Tool entwickelt, um Zielgene von ncRNAs vorherzusagen, wodurch 397 ncRNA-Ziel-Verbindungen in Mäusen und 2.291 in Menschen sowie deren Wirkungsweisen identifiziert wurden. |
